Evaluation of ethanolic extract of thyme leaves Thymus vulgaris as a virulence inhibitor in Vibrio parahaemolyticus

Abstract

Currently there are emerging diseases caused by bacteria, whose main problem is the early mortality syndrome transmitted by Vibrio parahaemolyticus causing mortality and economic losses in aquatic invertebrate culture. For this reason, the effectiveness of the extract of the Tomillo leaves Thymus vulgaris as a virulence inhibitor agent was investigated. In vitro assays of virulence factors such as minimum inhibitory concentration, cell density, mobility and biofilm were carried out. The results allowed determining inhibition in the bacterial proliferation of the tests at different concentrations, an effect that was perceived in the sensitivity for V. parahaemolyticus corresponding to the minimum inhibitory concentration at 3.5 mg/ l and 10.5 mg/ml. In the cell density, inhibitory action was observed at 17.5 mg/ml. In mobility tests in a semi-liquid medium (swimming) the bacteria decreased the ability to move to 17.5 mg/ml, 10.5 mg/ml and 3.5 mg/ml, as well as in a semi-solid medium (swarming) where the bacteria lacked the ability to move against all concentrations. Regarding biofilm formation, it was observed that the inhibitory effect results at 3.5 mg/ml. It is concluded that the thyme extract did not function as an antibacterial, but rather as a virulence inhibitor for V. parahaemolyticus, an action that is due to compounds present in the extract (indole and lactones). However, to establish its use in aquaculture, it is necessary to carry out purification tests of these two molecules to evaluate infections with Vibrio parahaemolyticus in shrimp larvae
