Introduction: Ovariohysterectomy is one of the most commonly performed surgical procedures in veterinary practice and is frequently associated with considerable pain. This pain is often inadequately managed, underscoring the importance of effective intraoperative and postoperative pain control strategies. Objective: To assess the efficacy of intraperitoneal instillation of two local anesthetics, in combination with dexmedetomidine, as adjuvants within a multimodal pain management protocol in bitches undergoing ovariohysterectomy. Methodology: Five treatment groups were established: T0 (control group), T1 (intraperitoneal lidocaine), T2 (intraperitoneal bupivacaine), T3 (combination of lidocaine and bupivacaine), and T4 (lidocaine plus intraperitoneal dexmedetomidine). Physiological and pharmacological parameters—including heart rate, respiratory rate, oxygen saturation, mean arterial pressure, capnography, body temperature, and Glasgow Composite Pain Scale scores—were assessed at 30 minutes, 8 hours, and 24 hours following surgery. Results: During surgery, pain was observed in 50% of patients in group T0 and 12.5% in T1, whereas 100% of animals in groups T2, T3, and T4 remained pain-free. Statistically significant differences (P < 0.05) were detected in pain scores over time. Groups receiving local anesthetics maintained effective analgesia without requiring rescue analgesia. Conclusion: Intraperitoneal instillation of local anesthetics is an effective strategy for controlling post-surgical pain in ovariohysterectomized bitches, contributing to improved animal welfare and recovery
